- Dendritic spines are numerous small protrusions on the dendrites of most neurons in the central nervous system. Each spine is connected with an axon terminal of a glutamatergic neuron, and contributes to the formation of an excitatory synapse as the postsynaptic compartment. Spines are dynamic structures that contain cytoskeletal, transmembrane and scaffolding proteins. The shapes and numbers of spines on a neuron vary widely and can be plastically altered by synaptic activity under physiological and pathological conditions. Dendritic spine plasticity has been thought to underlie fundamental brain functions such as learning and memory. In addition, various pediatric, neurological, and psychiatric disorders have elements of dendritic spine abnormalities. Spines become malformed or are lost in epilepsy, stroke, trauma, schizophrenia, dementia, major depression, normal aging, and chronic substance abuse. Because spine morphology is linked to synaptic function, altered spines in disease states are likely to have diverse functional effects. Recent advances in imaging and molecular technologies have revealed detailed information about physiological and pathological spine plasticity. This study reviews the latest advances in the field of activity-dependent spine remodeling and discusses the abnormalities in the development of brain diseases. © Nova Science Publishers, Inc.
