要  旨:DNA replication is a vital process in all living organisms. At each cell division > 30000 replication origins are activated in a coordinated manner to ensure the duplication of > 6 billion base pairs of the human genome. During differentiation and development this program must adapt to changes in chromatin organization and gene transcription: its deregulation can challenge genome stability which is a leading cause of many diseases including cancers and neurological disorders. Our team focuses on using high-throughput genomic approaches and genome-wide data analyses to study the spatio-temporal replication program of the human genome and its impact on genome stability.Thanks to these novel data we have revealed that gene transcription and chromatin organisation play important roles in shaping cell-type specific DNA replication landscape while transcription-mediated replication stresses also induce genome instability at both early and late replicating regions but via completely different mechanisms. More recently we have developed new high-throughput genome-wide approaches to study DNA replication at the single-molecule as well as single-cell level which allows studying DNA replication program in heterogeneous samples such as cancers.Recent key publications:
