The DNA polymerase epsilon (Polε) synthesizes the bulk of leading strand DNA. However, Polε is also responsible for the C-to-G mutations, the most prevalent mutations in cancers and genetic diseases. Polε has undergone a gene duplication event during evolution, leading to an N-terminal catalytic Pol domain and a C-terminal non-catalytic Pol domain. In my talk, I will discuss how such unusual architecture leads to a highly unique molecular mechanism of the enzyme in the leading strand replisome.
