Complex neural networks consist of many neurons with distinct identities. Though it is long appreciated that the enormous neural diversity arises in spatiotemporally patterned manners, the exact underlying molecular mechanisms remain unclear especially in mammalian brains. Mylab has developed and applied sophisticated genetic mosaic tools to understand how the Drosophila melanogaster brain develops on the level of individual cells. We have successfully mapped the developmental sequence of entire neuronal lineages in the adult fly brain, which provides unparalleled resolution and critical insight into how a limited number of progenitor cells can generate diverse neurons. We are keen to extend same approaches from fly to vertebrates (including mouse and zebrafish) to determine if conserved mechanisms govern the development of vertebrate brains. To this end, we have built TEMPO that allows tracking neuron birth order and manipulating gene functions acutely as desired. By TEMPO, we are reassessing the roles of Imp RNA-binding proteins in the development of mouse cortex. Determining the mechanisms of neural stem cell fates is crucial for understanding brain development & diseases and producing cell types at will for replacement therapy.
