Marco Foiani 教授は、染色体DNA損傷および複製ストレス障害の細胞応答の研究で世界的に著名で、Cell誌のEditorも務めておられます。今回の来日の機会に最新の研究成果についてご講演いただきます。
講演要旨
The advance of replication forks generates topological impediments that must be solved to warrant a correct replication process and to prevent aberrant DNA transitions leading to genome rearrangements and instability.
S phase transcription imposes tremendous topological stress to advancing forks and type II DNA topoisomerase organizes the architecture of transcription units to coordinate the clash between replication and transcription.
Replication termination also imposes topological stress to merging forks and specialized DNA helicases facilitate the termination process to prevent unscheduled events leading to chromosome fragmentation and rearrangements.
I will describe physiological and pathological events at replication forks dealing with transcription and with the termination problem.
